چکیده

Northebaine was identified as the major metabolite formed during the biotransformation of thebaine by Mucor piriformis. Minor metabolites identified are isomeric thebaine N-oxides. Among isomeric thebaine Noxides, the one with the equatorial oxygen attached to the nitrogen is relatively unstable. Resting cells. grown in the presence of thebaine for 24 hr, efficiently converted thebaine into northebaine (-77% conversion). Similar experiments carried out with northebaine and isomeric thebaine N-oxides as substrates, revealed that while northebaine and the stable thebaine N-oxide are proved to be resistant to further transformation, the unstable thebaine N-oxide nonenzymatically rearranges to 6, 7, 8, 9, 10, 14-hexadehydro-4, 5-epoxy-3. 6-dimethoxy-17methylthebinan and 6, 7, 8,9, 10, l4-hexadehydro-3. 6-dimethoxythebinan-4-01. These studies support the idea that N-demethylation of thebaine is hot proceeding via the N-oxide intermediate.

Urinary excretion of morphine and biosynthetic precursors in mice.

It has been firmly established that humans excrete a small but steady amount of the isoquinoline alkaloid morphine in their urine. It is unclear whether it is of dietary or endogenous origin. There is no doubt that a simple isoquinoline alkaloid, tetrahydropapaveroline (THP), is found in human and rodent brain as well as in human urine. This suggests a potential biogenetic relationship between ...

Behavioral effects of thebaine in the rhesus monkey.

Three experiments were conducted with rhesus monkeys to assess some behavioral effects of the opium alkaloid, thebaine, in relation to its dependence liability. The concurrent intramuscular administration of naloxone did not antagonize the rate-decreasing effects of thebaine on a fixed-ratio (FR) schedule of food-reinforced responding. Animals trained to self-administer codeine (0.3 mg/kg/inj) ...

Optimization of Thebaine Production using Adventitious Roots of Papaver bracteatum Lindle. by Alteration of Aeration Volume and Temperature in Bioreactor

Background: Nowadays, the optimization of the culture conditions in bioreactors is considered as an approach to produce secondary metabolites such as thebaine. Thebaine is the dominant alkaloid in Iranian poppy that is used as a precursor for the synthesis of analgesic compounds. Objective: Optimization of culture conditions of Papaver bracteatum adventitious roots in bioreactor for the scale...

Megistolactone, a new alkaloid from Sarcomelicope megistophylla.

A new quinolone alkaloid, megistolactone (1) was isolated from the bark of Sarcomelicope megistophylla. Its structure has been elucidated on the basis of MS and NMR data. From a biogenetic point of view, this compound should be considered as an oxidation product of 1,2,3,4-tetra-O-subsituted acridone alkaloids, which are also present in the bark.

How human neuroblastoma cells make morphine.

Recently, our laboratory demonstrated that human neuroblastoma cells (SH-SY5Y) are capable of synthesizing morphine, the major active metabolite of opium poppy. Now our experiments are further substantiated by extending the biochemical studies to the entire morphine pathway in this human cell line. L-[1,2,3-13C3]- and [ring-2',5',6'-2H3]dopa showed high isotopic enrichment and incorporation in ...
